This multi-perspective study on an implementation   process of portfolios in an Austrian Gymnasium   context stretches over more than three years. The   dichotomy caused by implementing and using   portfolios as learning strategy and alternative   assessment tool in a fairly traditional educational   context is the central research focus. Process   details as well as outcomes are critically looked   upon from the perspectives of the learners, the   teacher and the contextual setting.

<p>Liquids under the influence of external fields exhibit a wide range of intriguing phenomena that can be markedly different from the behaviour of a quiescent system. This work considers two different systems - a glassforming Yukawa system and a colloid-polymer mixture - by Molecular Dynamics computer simulations. The former consists of a 50-50 binary mixture of differently-sized, like-charged colloids interacting via a screened Coulomb (Yukawa) potential. Near the glass transition the influence of an external shear field is studied. In particular, the transition from elastic response to plastic flow is of interest. The obtained results are important for advancing new theoretical approaches in the framework of the mode-coupling theory of the glass transition. Furthermore, they suggest new experimental investigations on colloidal systems. As second system a colloid-polymer mixture is studied in the context of its behaviour near the critical point of phase separation. Based on the Asakura-Oosawa model a new effective model with soft interaction potentials is introduced and characterised in equilibrium. Finally its suitability for shear simulations is shown.</p>

The majority of ATP, the carrier of energy in the living world, is generated by the ATP synthase. The enzyme complex converts energy from the transmembrane electrochemical gradient into rotation by an H+ or Na+ current through the F0 part. The rotation is transmitted to the F1 part, where ATP is formed from ADP and Pi. The present thesis aimed to elucidate the requirements for torque generation during ATP synthesis in the holoenzyme as well as in the isolated F0 part. To achieve this, a quantitative H+ transport assay was established. Furthermore, the Na+-dependent enzyme from P. modestum and the H+- dependent ATP synthase from E. coli were compared with respect to torque generation and ATP synthesis. We observed that a small ion concentration gradient was indispensable for ATP synthesis in both enzymes. Much more surprising was the observation that ~ 100 times higher Na+ or H+ concentrations were required for synthesis than hydrolysis of ATP. For the first time the ion concentration at the periplasmic side was recognized as critical determinant for ATP synthesis. Based on these data an model the F-type ATPases is presented.

In this thesis (R)-3-hydroxycarboxylic acids (HA) from bacterial polyhydroxyalkanoates (PHA) were investigated, regarding their biotechnological production as well as intracellular processes of their formation and degradation. An innovative bioprocess to provide HA as fine chemicals for further applications is presented. The method is straightforward, easy to handle and results in high yields (introduced in chapters 2-4). Following the observations of this study, attempts to scale up the process seemed possible and potential applications of HA remain topics of research. The underlying mechanisms of PHA degradation were also addressed, including issues of physiological advantages of HA excretion for cells (chapter 5) and characterization of enzymes having HA as substrates (chapters 6 and 7). Even though some aspects of PHA metabolism were clarified for P. putida GPo1, many details about intracellular PHA degradation remain obscure and require further experiments and discussion. Knowledge about their molecular context in the bacterial cell is crucial for efficient PHA and HA production.

In this thesis, the influence of noise, variability, and time-delayed feedback on the dynamics of nets of neural elements is investigated. The dynamics of the single elements is given by the FitzHugh-Nagumo, the Hodgkin-Huxley, or the reduced Hodgkin-Huxley equations. After some theoretical basics, the results are presented. For oscillatory nets, it is shown that time-delayed feedback can suppress oscillations and thereby excitable net dynamics is induced. In subexcitable nets, time-delayed feedback can support the propagation of wave fronts and thereby pattern formation. This effect is studied in detail for wave fronts, which are either induced by special initial conditions, by noise, or by variability. For bistable nets, it is shown that both multiplicative noise and multiplicative variability have a systematic influence on the net dynamics. Considering an external signal, the response of the net is optimal for intermediate values of the additive and the multiplicative variability strength (doubly variability-induced resonance). Moreover, the response of the net to the external signal can further be enhanced applying time-delayed feedback with appropriately chosen delay times.

Computational Fluid Dynamics (CFD) simulations of large-scale JP-4 pool fires in calm and cross-wind conditions were performed to predict emission temperatures, surface emissive power, and irradiances. CFD simulations were done in a one million cell computation mesh by using the Scale Adaptive Simulation and Large Eddy Simulation including flamelet models with 20-112 species and 60-800 chemical reactions. This work presents three new ways to predict surface emissive power by CFD. To simulate the surface emissive power a four-step discontinuity function consisting of a temperature dependent effective absorption coefficient of the dissipative structures (reaction zones, hot spots and soot parcels) and air was developed. Instantaneous and time averaged thermograms were used to determine the probability density functions of the emission surface temperatures and the surface emissive power. CFD simulations showed how the wind influenced the flame length, flame tilt, flame drag, flame temperatures, surface emissive power, and irradiances. Using these CFD simulations the thermal radiation from large pool fires can be more accurately estimated.
